Pregnil 5000 is a drug with a gonodotropic effect, produced in the form of a white lyophilisate (powder) for the preparation of an injection solution that is administered subcutaneously or intramuscularly.

The package with the medicine contains a solvent that looks like a colorless, completely transparent liquid. The solvent is packaged in ampoules of 2 ml, blister packs and cardboard boxes. The medicine kit includes 3 ampoules of solvent.

Ampoules contain different concentrations of human chorionic gonadotropin - 500 IU, 1500 IU, 5000 IU. The solvent contains 0.9% sodium chloride solution.

The price for the drug in question varies, it all depends on how many pieces are in the package, as well as on the pharmacy where it is purchased. The average price varies by:

  • Pregnil 5000 IU - costs from 250 to 450 rubles. The package contains two ampoules for one injection;
  • Pregnil 1500 IU - found in pharmacy chains at an average cost of 1200 rubles, the drug is enough for three uses;

Pregnil 5000 refers to pharmacological drugs with gonadotropic action. The biological activity of hCG, which is part of it, is completely identical to the action of luteinizing hormone, which takes part in the formation of male and female sex hormones and gametes (reproductive cells).

The use of the drug Pregnil 5000 for women is aimed at replacing the release of endogenous luteinizing hormone in the middle phase of the cycle. This stimulates and accelerates the onset of the final stage of follicle maturation. As a result, ovulation occurs.

In some cases, the drug is prescribed during the luteal phase as an analogue of luteinizing hormone.

The use of the pharmaceutical drug in men and boys is recommended if necessary to stimulate Leydig cells that produce testosterone.

Efficiency

According to experts, this medicinal substance is one of the most effective injections for inducing ovulation. It was created from human chorionic gonadotropin. Due to luteotropin, the growth of male hormones (steroids) in the girl’s reproductive organs is provoked. But it is worth considering that a reverse reaction of the body is possible - the appearance of antibodies. This is one of the reasons why you should not self-medicate.

The drug is not suitable for everyone, as it has a very narrow range of properties:

  • provoking follicle growth, stimulating ovulation if it is absent;
  • ensuring proper development for the follicle;
  • assistance in the third phase of the menstrual cycle.

Therefore, if a woman cannot give birth due to problems with the fallopian tubes, the injections will be pointless.

Pharmacodynamics of the drug

Pregnyl 5000 is a natural hormone human chorionic gonadotropin, which is produced by the human placenta. This substance enters the urine, from where it is extracted and purified. Action of the drug:

  • activates the production of the hormone progesterone by the corpus luteum;
  • ensures the full development of the placenta;
  • stimulates ovulation, accelerates the synthesis of progesterone and estrogen;
  • activates the production of testosterone in the body;
  • in men – activates the production of sex steroids and improves spermatogenesis, helping to fight infertility.

Pregnil 5000 is intended for injection - intramuscular or subcutaneous. The pharmaceutical product promotes increased production of steroid hormones in the male and female gonads. When this hormone is used, the body does not develop the process of producing antibodies to it.

The maximum concentration of the active substance in the blood plasma of women is observed 19–20 hours after the hCG injection. For men, this figure is 7–16 hours. The difference in the rate of absorption of human chorionic gonadotropin is associated with denser fatty tissue in women.

Metabolism of pharmaceuticals occurs in the liver. The half-life of Pregnil 5000 from the body is 32–33 hours. The drug is excreted naturally by the kidneys, approximately 10% of the used dosage every 24 hours.

In the case of systematic use of a gonadotropic drug, a gradual increase in the level of human chorionic gonadotropin in human blood plasma is observed. The instructions for the drug Pregnil 5000 indicate all the indications for its use. Recommendations for women:

  • support needed for corpus luteum deficiency;
  • dysmenorrhea, ovarian dysfunction;
  • infertility caused by lack of ovulation or disruption of the maturation of follicles;
  • as an element of a program of controlled ovarian hyperstimulation (during the preparation of follicles for puncture and assisted reproductive techniques).

Pregnil 5000 injections are prescribed for recurrent miscarriage or a high risk of spontaneous miscarriage.

Human chorionic gonadotropin is also used for men, boys and adolescents. Main indications for use:

  • diagnosing cryptorchidism not associated with anatomical obstruction;
  • the presence of hypogonadotropic hypogonadism;
  • delayed puberty due to insufficiency of gonadotropic functions of the pituitary gland;

Contraindications

Pregnil 5000 has a list of contraindications for use. Intramuscular or subcutaneous injection of hCG is prohibited in the following cases:

  1. For male teenagers – premature puberty.
  2. In men, prostate cancer.
  3. In women, malignant neoplasms of the hormone-dependent type are uterine cancer, ovarian or mammary gland cancer.
  4. Fibrous uterine tumors incompatible with pregnancy.
  5. Anatomical pathologies of the reproductive organs incompatible with pregnancy.

The drug is prohibited to be used throughout the entire period of pregnancy and breastfeeding, as well as in case of individual intolerance to the active or auxiliary components included in its composition.

The instructions for the medication highlight a number of relative contraindications, in which the use of Pregnil 5000 is carried out with extreme caution and exclusively under strict medical supervision.

  1. In women: obesity, increased risk of thrombosis and thrombophilia, venous or arterial thromboembolism during treatment with gonadotropins.
  2. Men and adolescents - hypertension, overt or latent heart failure, kidney dysfunction, epilepsy, frequent migraines.
  3. Boys are a prepubertal period of development (high levels of hCG can trigger premature puberty).

Pregnyl can cause side effects in the patient's body with varying degrees of severity.

Side effects in women:

  • development of ovarian hyperstimulation syndrome, accompanied by pain in the lower abdomen and mammary glands, the formation of cysts in the ovaries and an increase in their size;
  • pain in the mammary glands;
  • painful cramps in the stomach, nausea, upset stool;
  • headache;
  • thromboembolism;
  • rapid weight gain;
  • increased body temperature, fever;
  • acne on the skin;
  • sudden changes in mood, nervousness, irritability, anxiety;
  • development of chronic fatigue, depressive states.

If ovarian hyperstimulation syndrome develops, you must stop using the drug and avoid pregnancy. The syndrome can occur in a severe form, accompanied by ascites, a sharp increase in body weight, and the formation of large cysts in the ovaries that are prone to rupture. This condition poses a serious danger to a woman’s life.

Frequent side effects of gonadotropin can be expressed in the form of redness, swelling, swelling, or bruising at the injection site. Local allergic reactions are possible, accompanied by skin rashes, itching and burning at the injection site. In the most severe cases, angioedema develops.

The use of Pregnil 5000 among boys and men is accompanied by increased formation of androgens, which results in fluid and sodium retention in the body. Also in male patients, enlargement of the prostate and mammary glands is possible, which is reversible.

How to prick

Before administering the drug, you must add sodium chloride to it using a 2 mm needle and syringe (it is in the package).

Mixing process

The needle pack contains large pink or orange needles for mixing and small ones for injecting.

  1. Place the yellow or pink needle on the syringe and remove the cap.
  2. Then pick up the ampoule with the solution and check if there is any water on its spout. If there is one, tap the tips with your finger to force the water down.
  3. Place your thumb on the indication line on the spout and break it off with a quick movement of your hand. Use a napkin to avoid cutting yourself.
  4. Place the sodium chloride ampoule on a flat surface, insert the syringe into it and pull the plunger back completely to absorb all the liquid. This is easier to do if you carry out the manipulation at a slight angle.
  5. In the same way, break off the spout of the second ampoule, slowly inject into it the sodium chloride that you collected into the syringe. To remove powder that has stuck to the sides of the ampoule and therefore has not dissolved, gently shake it, but do not shake it.
  6. Without removing the needle from the ampoule, move it back and forth a little, then pull back the plunger so that all the medicine enters. You have prepared a dose of 5,000 ppm.

After this, give an injection intramuscularly. An important point is that the procedure must be carried out slowly. Do not store the solution for a long time, because it will become unsterile and therefore unusable.

Compatibility with other drugs and alcohol

To date, in pharmacological practice there is no information on the interaction of human chorionic gonadotropin with other drugs.

The instructions for the medication emphasize that for 10 days before and after completion of therapy, the level of human chorionic gonadotropin increases significantly. This may cause a false positive result during a pregnancy test.

During the course of treatment with Pregnil 5000, the consumption of alcoholic beverages is prohibited. This significantly increases the likelihood of developing side effects of the medication.

Analogs

The presented drug has quite a few analogues, some of the popular ones are:

  • Utrozhestan;
  • Ovitrel;
  • Duphaston;
  • Chorionic Gonadotropin.

All of the above analogues are similar in their action. Therefore, it is not possible to say which has a better effect, for example Ovitrel or Regnil.

Use of the drug

To prepare the injection solution Pregnil 5000, intended for intramuscular or subcutaneous administration, the powdered lyophilisate must be mixed with a solvent.

After opening the ampoule and solvent, the prepared solution must be used immediately; it cannot be stored further due to loss of sterility.

The method of use of the drug and the recommended dosage of the drug depend on the gender, age of the patient, as well as the course of the disease.

For women:

  1. Infertility due to lack of ovulation or disruption in the process of follicle maturation - 1 injection with a concentration of 5000 IU.
  2. Preparation for assisted reproductive techniques or puncture if ovarian hyperstimulation is necessary - 1 injection with a concentration of 5000–10,000 IU.
  3. Insufficiency of the corpus luteum - two or three repeated injections on the 3rd, 6th or 9th day after ovulation.

For men and boys, the recommended dosage depends on the age of the patient and the type of disease. The average duration of treatment for infertility or delayed puberty is 3–6 months.

Application for men:

  1. For hypogonadism of the hypogonadotropic type - 2-3 injections over 7 days with a concentration of 1000-2000 IU.
  2. Delayed puberty due to insufficiency of gonadotropic functions of the pituitary gland - 2-3 injections with a dosage of 1500 IU over the course of a week.
  3. For cryptorchidism - 2 injections over the course of a week with a dosage of 1500 IU for boys over 6 years old, 500-1000 IU for 2-6 years old and 250 IU for boys under 2 years old.

Pregnil 5000 is used in the complex treatment of male infertility. The recommended dosage is 1000–2000 IU of the drug 2–3 times a week. In most cases, improvement in spermatogenesis occurs within 10–12 weeks from the start of treatment.

  3. Pregnil 5000 can be used to treat boys. The exception is premature puberty. During the prepubertal period, the drug is used with extreme caution, as it can provoke closure of the epiphyses and premature puberty.
  4. When treated with gonadotropin, patients with impaired renal function should be under constant medical supervision, since increased production of androgens can cause exacerbation or relapse of renal disease.
  5. Chorionic gonadotropin is not recommended for long-term use, as this increases the likelihood of the production of specific antibodies in the body, which reduce the effectiveness of therapy.

In case of hypersensitivity to human chorionic gonadotropin, it is recommended to discontinue Pregnil 5000 and select a drug with a similar principle of action. I’ll say right away that Pregnil is a good thing. The first time I couldn’t get pregnant for about two years. The doctors suffered with me for a long time. They monitored my body processes many times and did multiple tests. In conclusion, they said that I have very rare ovulation and the ovulation process itself is difficult. I can explain it differently: my mature egg could not come out. It simply degenerated over time, decreased, and in the end simply dissolved. The drug helped me then. But I would like to note that Pregnil is still a hormonal drug. Therefore, it has side effects that occur. I consider ovarian hyperstimulation to be the most dangerous. Therefore, I was a supporter of constant ultrasound examination. The doctors tried their best, they carefully monitored my processes in the body and their individual course. The drug helped me well with ovulation. She was as expected. If we talk about my feelings, I tolerated the drug itself well. However, the injections hurt a lot. They are painful. It felt like I was being cut with hot glass. I even had redness at the injection site, fortunately there were no lumps. I want to say that Pregnil works, but this is a very serious drug that must be taken under the supervision of specialists. I recommend Pregnil to all girls who have problems.
