In the article we will look at the instructions for use of “Proserin” injections (intramuscular).

This is a synthetic type cholinesterase inhibitor, an effective and well-studied drug. This medication is one of the most important and necessary for human life. When properly prescribed, it works great and helps well in solving a wide range of different problems in the neurological field.

Despite many positive results, it is prohibited to use the drug without preliminary diagnosis and medical consultation. Proserin has quite a few side effects and contraindications for use.

pharmachologic effect

The pharmacological group of the drug is a cholinesterase inhibitor. This is a synthetic anticholinesterase drug that reversibly blocks cholinesterase. As a result of this action, acetylcholine accumulates and its effect on tissues and organs increases. As a result, neuromuscular conduction is restored.

Under the influence of the drug, the heart rate decreases, the secretion of the bronchial, salivary, sweat glands, as well as the glands of the digestive tract increases. In turn, this contributes to the manifestation of bronchorrhea, hypersalivation, and an increase in the level of acidity of gastric juice.

The active substance causes a spasm of accommodation , constriction of the pupil, lowers intraocular pressure , stimulates the tone of the smooth muscles of the intestines and bladder, tones the skeletal muscles, and provokes bronchospasm.

Prozerin is an antagonist of anti-depolarizing curare-like drugs. When taking large doses, the drug can provoke a disorder of neuromuscular conduction, which occurs due to the accumulation of acetylcholine. Produces a direct n-cholinomimetic effect. The effect of the drug coincides with the characteristic effects of excitation of cholinergic nerves. It does not cause central effects; compared to physostigmine, it provides more active stimulation of the muscles of the bladder, intestines, and uterus.
The effect on the heart is minor.

In childhood

The medicine can be prescribed to children of any age, but the dosage prescribed by the doctor must be strictly followed. Children under 10 years of age are prescribed 1 mg of the drug (suspension) per day. For adolescents over 10 years of age, the daily dosage is up to 10 mg. Proserin injections are prescribed to children under 12 years of age at 0.2-0.5 mg. The administration regimen and duration of treatment are determined individually by the attending physician. You should know that Proserinum for children in all dosage forms is prescribed only in a hospital setting.

Indications for use of Proserin

There are the following indications for the use of Proserin:

  • myasthenia gravis;
  • paralysis;
  • movement disorders after brain injury;

The use of the product for children is practiced in similar cases.

Contraindications

  • other manifestations: increased urination, increased sweating, arthralgia.

Instructions for use of Proserin (Method and dosage)

Prozerin injections, instructions for use

The solution contained in the ampoules is used for subcutaneous administration in a dose of 1 ml 1-2 times a day. The largest single dose for subcutaneous administration is 0.002 g, per day - 0.006 g.

In order to use the medicine to stop the effect of muscle relaxants, atropine sulfate (0.5-0.7 mg) is initially administered intravenously, then wait until the pulse increases and after about 2 minutes 1.5 mg of Proserin is administered intravenously. If the effect is insufficient, a similar dose is re-administered. A total of 5-6 mg of the drug is allowed to be administered over about 30 minutes.

In ophthalmology, it is practiced to administer 1-2 drops into the conjunctival sac 1 to 4 times a day.

Oral administration is carried out at the rate of 10-15 mg 2-3 times a day for adults and 1 mg per day for 1 year of life for children under 10 years of age. The maximum permissible dose for adults is 50 mg per day, for children - 10 mg per day.

Reviews

Proserin is a well-studied and inexpensive drug; it has a good reputation among doctors, and therefore the solution is often prescribed to patients.

People note the following advantages in reviews of Proserin injections: affordable cost and high effectiveness, absence of complications and problems after treatment.

Other patients say that you need to be extremely careful with the medication, as it is not suitable for everyone.

Disadvantages include painful injections and, in some cases, difficult tolerability. Sometimes severe dizziness and nausea occur. The drug is very powerful and, if used incorrectly, can harm the patient. However, when prescribed correctly, it is effective and helps people perfectly.

Overdose

An overdose of the drug causes symptoms that are associated with the process of overexcitation of cholinergic receptors (the so-called cholinergic crisis). Hypersalivation, miosis, bradycardia, nausea, bronchospasm, diarrhea , frequent urination, general weakness, twitching of skeletal muscles and tongue, and decreased blood pressure .

In this case, you need to reduce the dose of the medicine or stop taking it completely. If there is such a need, the patient is administered Atropine , Metacin , as well as other drugs with an anticholinergic effect.

How does it work?

Electrophoresis is the effect of direct electric current on body tissue. At the same time, a medicinal substance is supplied from the electrode, which breaks down into ions. They penetrate the tissue and have the pharmacological effect characteristic of the drug. For each drug, the concentration is selected and the pole from which it will be administered is determined. For Proserin this is a 0.1% solution from the anode.

The action of the drug and the physiotherapeutic effect are summed up, which allows you to achieve great results. The resulting effect is determined by the mobility of particles in the field created by the device, as well as the concentration used and the area of influence.

The closer to the pathological process the procedure is performed, the better the technique works.

The percentage of the drug that penetrates tissue during electrophoresis does not exceed 10%. Scientists conducted an experiment that showed the inconsistency of the theory that a high concentration of the drug has a positive effect on the effectiveness of the procedure. This is due to the inclusion of ion braking forces for solutions of 5% and higher. It is called the Debye-Hückel phenomenon.

Due to the low permeability of solutions during electrophoresis, only strong drugs or electrolyte solutions are used for this method. Under the influence of current, drugs change, which adds new properties to them or enhances existing ones. Local exposure to the device ensures point accumulation of the active substance. This helps to avoid significant side effects that occur with systemic use.

Interaction

It should be borne in mind that neostigmine methyl sulfate is an antagonist of non-depolarizing muscle relaxants, therefore, when taken simultaneously, it activates the effect of depolarizing muscle relaxants.

If ganglion blockers, m-anticholinergic blockers, Novocainamide, Quinidine, local anesthetics antidepressants , drugs with antiepileptic and antiparkinsonian effects are used simultaneously, the effect of neostigmine methyl sulfate is reduced.

The effect of neostigmine methyl sulfate is potentiated when taking Ephedrine .

When taking neostigmine methylsulfate and beta-blockers, bradycardia may increase .

You are absolutely right, Stas.ReplyReply

I’m not allergic, but the reaction is unpleasant, it goes away quickly, I would have endured it since I needed it so much, but the doctor canceled it, it wasn’t enough, he says that you should move my horses here. So who cares ReplyReply

Nata, Prozerin is indeed used for diseases that are accompanied by the symptoms you described. You will agree that it is impossible to judge the correctness of the prescription remotely, without even knowing the diagnosis. In a situation where the side effect is so pronounced, you need to stop taking the drug and consult your doctor about this, he will either adjust the dose or select another drug. If it is not possible to get an appointment with him, try contacting him by phone. Reply

special instructions

Injections and tablets are used carefully for people with Addison's disease.

When administering the drug parenterally in large doses, Atropine must be administered simultaneously or pre-administered.

If during treatment a myasthenic crisis (due to an insufficient dose) or a cholinergic crisis (due to an overdose) develops, a careful differential diagnosis must be carried out due to the similarity of symptoms.

It is recommended to refrain from driving or engaging in other hazardous activities that require maximum concentration.
